Currently, there is a limited number of studies on the state of platelet hemostasis in children with ET.</p> <p><bold>Aim</bold> of this single-center prospective study was to characterize the morphological and functional properties of platelets in pediatric patients with ET.</p> <p><bold>Materials and methods. </bold>The study included 30 patients (10 boys and 20 girls) aged 3–17 years with an established diagnosis of ET. The control group consisted of 45 healthy volunteers (22 boys and 23 girls) aged 4–18 years. Flow cytometry with strong and weak activation, aggregometry, and immunofluorescence microscopy of blood smears were used to analyze platelet function.</p> <p><bold>Results.</bold> It was shown that platelets of patients with ET are characterized by a reduced size, while the number of platelet alpha and dense granules did not differ from the control group. Platelets of patients with ET showed moderate hyporeactivity in response to strong stimulation (by activation of glycoprotein IIbIIIa and secretion of dense granules). Platelet aggregation with collagen and adenosine diphosphate was also reduced in patients with ET, but these data did not correlate with the degree of platelet activation in flow cytometry.</p> <p><bold>Conclusion.</bold> Thus, characteristic morphological and functional changes in platelets were revealed in children with ET compared with the control group.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> Эссенциальная тромбоцитемия (ЭТ) – орфанное миелопролиферативное заболевание с избыточной выработкой тромбоцитов, ассоциированное с повышенным риском тромбозов и кровотечений. В настоящее время существует ограниченное количество исследований, посвященных состоянию тромбоцитарного звена гемостаза у детей с ЭТ.</p> <p><bold>Цель исследования </bold>– охарактеризовать морфологические и функциональные свойства тромбоцитов у педиатрических пациентов с ЭТ.</p> <p><bold>Материалы и методы.</bold> В одноцентровое проспективное исследование были включены 30 пациентов (10 мальчиков и 20 девочек) в возрасте 3–17 лет с установленным диагнозом ЭТ. Контрольную группу составили 45 здоровых добровольцев (22 мальчика и 23 девочки) в возрасте 4–18 лет. Для анализа функции тромбоцитов использовали проточную цитометрию с сильной и слабой активацией, агрегометрию и иммунофлуоресцентную микроскопию мазков крови.</p> <p><bold>Результаты.</bold> Показано, что тромбоциты пациентов с ЭТ характеризуются уменьшенным размером, при этом количество тромбоцитарных альфа- и плотных гранул не отличалось от контрольной группы. Тромбоциты пациентов с ЭТ проявляли умеренную гипореактивность в ответ на сильную cтимуляцию (по активации гликопротеина IIbIIIa и секреции плотных гранул).
