Despite advances in understanding its pathogenesis, treatment options are still limited, and a significant proportion of patients remain transfusion-dependent.</p> <p><bold>Aim:</bold> to evaluate the effect of eltrombopag on the colony-forming potential of bone marrow erythroid progenitors in DBA patients <italic>in vitro</italic>.</p> <p><bold>Materials and methods.</bold> The study included 10 patients with genetically confirmed DBA and 7 healthy donors. Bone marrow mononuclear cells were cultured in a methylcellulose medium supplemented with eltrombopag at concentrations of 13 and 26 μg/mL.</p> <p><bold>Results.</bold> <italic>In vitro</italic>, the addition of eltrombopag to bone marrow cell cultures from the patients with DBA resulted in a significant increase in the number of erythroid colonies compared to control cultures without the drug. For early erythroid progenitors, a statistically significant increase in the number of colonies was observed.</p> <p><bold>Conclusion.</bold> These interim results suggest that eltrombopag may be considered a potential stimulator of erythropoiesis in DBA and support the need for further research in larger cohorts in order to clarify the clinical significance of the observed effects.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> Анемия Даймонда–Блекфена (АДБ) – редкий врожденный синдром костномозговой недостаточности, относящийся к группе рибосомопатий и характеризующийся селективной аплазией эритроидного ростка гемопоэза. Несмотря на прогресс, достигнутый в понимании патогенеза, терапевтические возможности остаются ограниченными, а значительная доля пациентов сохраняет трансфузионную зависимость.</p> <p><bold>Цель исследования</bold> – оценка влияния элтромбопага на колониеобразующий потенциал эритроидных предшественников костного мозга у пациентов с АДБ<italic> </italic><italic>in</italic><italic> </italic><italic>vitro</italic>.</p> <p><bold>Материалы и методы.</bold> В исследование были включены 10 пациентов с генетически верифицированной АДБ и 7 здоровых доноров. Мононуклеарные клетки костного мозга культивировали в метилцеллюлозной среде с добавлением элтромбопага в концентрациях 13 и 26 мкг/мл.</p> <p><bold>Результаты.</bold> В условиях<italic> </italic><italic>in</italic><italic> </italic><italic>vitro</italic> добавление элтромбопага в культуру клеток костного мозга пациентов с АДБ сопровождалось увеличением числа эритроидных колоний по сравнению с контрольными культурами без добавления препарата.
